Pre-clinical mechanism
Binds the pituitary GHRH receptor, stimulating endogenous growth hormone release through a pulsatile pattern resembling physiological GH secretion. The DAC (drug affinity complex) variant carries a maleimide group that reacts with serum albumin in animal models, extending plasma half-life.
Key pre-clinical findings
- Rodent and canine pharmacokinetic studies: extended half-life of the DAC variant relative to unmodified GHRH (Teichman et al., 2006).
- Healthy adult phase-1 pharmacology: dose-dependent increases in GH and IGF-1 with weekly administration (Teichman et al., 2006).
