Pre-clinical mechanism
Selective agonist at the glucagon-like peptide-1 receptor. Pre-clinical and clinical literature describes glucose-dependent insulin secretion, suppression of glucagon release, slowed gastric emptying and central nervous system effects on satiety.
Key pre-clinical findings
- STEP-1 trial (adults with obesity, 68 weeks): mean body weight change of -14.9% with semaglutide vs -2.4% with placebo (Wilding et al., 2021).
- SUSTAIN-6 cardiovascular outcome trial: reduction in composite cardiovascular endpoints in adults with type-2 diabetes (Marso et al., 2016).
