Pre-clinical mechanism
Converted in liver and kidney to the active hormone calcitriol (1,25-(OH)₂D), which acts via the nuclear vitamin D receptor (VDR) to regulate calcium-phosphate handling, immune-cell differentiation and hundreds of downstream genes.
Key pre-clinical findings
- Endocrine Society clinical-practice review summarises current evidence on 25(OH)D status and supplementation (Holick et al., 2011).
